- Abstract
- The endoplasmic reticulum (ER) serves as a central hub of the intracellular membrane network, interacting with diverse organelles through membrane contact sites. Atlastins (ATLs) are dynamin-like GTPases that mediate homotypic membrane fusion between ER tubules and play a critical role in the formation and maintenance of three-way junctions within the ER network. Mammals express three ATL paralogs. ATL1, which is predominantly expressed in neurons, has been implicated in endosomal fission and lysosomal protein degradation in neuronal cells. However, the impact of ER network integrity on endo-lysosomal homeostasis in non-neuronal cells remains poorly understood. In this study, I examined the effects of ATL2/3 depletion on endo-lysosomal morphology and function in non-neuronal cells. Loss of ATL2/3 resulted in marked enlargement of early endosomes and lysosomes. These morphological changes were restored to near-WT levels by re-expression of ATL2. Beyond morphological abnormalities, ATL2/3 depletion caused widespread lysosomal dysfunction. In particular, ATL2/3-depleted cells showed defective lysosomal acidification, accompanied by cholesterol accumulation within lysosomes and reduced lysosomal protease activity. Although transferrin recycling was not significantly altered by ATL2/3 loss, the intracellular fluorescence signal of EGF — a canonical cargo of the degradative pathway — declined more rapidly in ATL2/3-depleted cells. Collectively, these findings demonstrate that the ER-shaping proteins ATLs play an important role in regulating endo-lysosomal homeostasis. Thus, this study may further provide insight into how ATLs contribute to endo-lysosomal dysfunction associated with ATL-related disease contexts.|소포체(endoplasmic reticulum, ER)은 세포 내 막 네트워크의 중심 허브로서, 막 접촉 부위를 통해 다양한 세포소기관과 상호작용한다. Atlastins(ATLs)는 ER 세관 간의 동종 막 융합을 매개하는 dynamin-유사 GTPase로, ER 네트워크 내 삼방향 접합부(three-way junction)의 형성과 유지에 핵심적인 역할을 한다. 포유류는 세 가지 ATL paralogs을 발현한다. 주로 신경세포에 발현하는 ATL1이 뉴런에서 엔도솜 분열과 리소좀 단백질 분해능에 관여함이 밝혀졌다. 그러나 비신경세포에서 ER 네트워크의 구조적 완전성이 엔도-리소좀 항상성에 미치는 영향은 아직 충분히 되지 않았다. 본 연구에서는 비신경세포에서 ATL2/3 결핍이 엔도-리소좀의 형태와 기능에 미치는 영향을 분석하였다. ATL2/3가 결핍되면 초기 엔도솜과 리소좀이 현저히 비대화되었다. 이러한 형태적 결함은 ATL2를 재발현시켰을 때 야생형 수준에 가깝게 회복되었다. 형태적 이상뿐만 아니라, ATL2/3 결핍은 전반적인 리소좀 기능 이상을 초래하였다. 특히 ATL2/3 결핍 세포에서는 리소좀 산성화 저하와 함께 콜레스테롤이 리소좀 내에 축적되었으며, 리소좀 단백질분해효소 활성 또한 감소하였다. Transferrin 재활용은 ATL2/3 결핍에 의해 유의하게 변화하지 않았으나, 분해 경로의 대표적인 화물인 EGF의 세포 내 형광 신호는 ATL2/3 결핍 세포에서 더 빠르게 감소하였다. 종합하면, 본 연구 결과는 ER 형태 조절 단백질인 ATL이 엔도-리소좀 항상성 조절에 중요한 역할을 한다는 것을 보여준다. 따라서 본 연구는 ATL이 ATL 관련 질환 맥락에서 나타나는 엔도-리소좀 기능 이상에 어떻게 기여하는지에 대한 이해를 넓히는 데 도움을 줄 수 있다.
