A role of diuretic hormone 44 receptor 1 in the acoustically primed sexual behavior

Abstract

초파리 암컷은 자신의 생리적인 상태 등 내적 상태 그리고 수컷으로부터 오는 시각, 청각, 후각과 같은 다양한 외부의 단서에 의해 교미여부를 결정한다. 그 중 청각적인 단서인 수컷의 구애 노래 는 암컷 초파리를 흥분시켜 교미를 유도하는 역할을 한다고 알려져 있다. 구애 노래는 일반적으 로 암컷을 성적으로 흥분시키는 요소로 알려져 있지만, 구애 노래에 미리 노출될 경우에는 다음 에 있을 교미행동에 영향을 주어 교미에 걸리는 시간을 짧아지게 할 수 있다는 연구가 보고된 바 있다. 이러한 현상은 성적 프라이밍 효과라고 알려졌는데, 이는 다른 모델동물인 설치류에서 역시 나타나는 현상으로 알려져 있습니다. 청각 자극의 선행경험으로 인한 프라이밍 효과를 측정하기 위해 야생형인 Canton-S를 이용하여 행동실험기법을 최적화했습니다. 수컷 구애 노래 중 sine song을 인식하여 교미행동을 변화시키는데 관여하는 인터페이스 뉴런 중 하나로 pC1 뉴런이 있 다. pC1 뉴런은 암컷 뇌에서 특이적이로 발현하는 뉴런으로 수컷으로부터 오는 구애노래에 반응 하여 칼슘 반응을 보이고 암컷의 교미결정에 관여하는 뉴런으로 보고되었다. pC1 뉴런에 특이적 으로 발현하는 pC1-GAL4를 이용하여 sine song에 의한 반응을 관찰해본 결과 cAMP의 양이 높아 지는 것을 확인하였다. 또한 pC1에 있는 Dh44의 수용체 Dh44R1의 발현을 RNAi로 저해했을 경 우, sine song을 노출시켜 주어도 cAMP의 양이 변하지 않는 현상이 나타났다. 이 결과는 수컷의 sine song이 암컷의 성적 프라이밍을 유도하며, 이때 pC1 세포에서 작용하는 Dh44R1 신호전달 시스템이 작용하고 있음을 제시한다|Female Drosophila makes mating decisions based on her internal state and male courtship with visual, auditory and olfactory cues. The male courtship songs consisting of the sine and pulse song are essential for female mating. In particular, sine song component arouses females sexually and makes them engaged in mating with shorter lantency. This phenomenon was known as the sexual priming effect in Drosophila, and a similar phenomenon was also observed in rodents. Here, I attempted to establish a behavioral protocol that measures the acoustic priming effect of sine song. I found pC1 neurons that regulate female receptivity and show Ca2+ responses to the courtship song show a marked cAMP increase in response to sine song but not to pulse song. Knockdown of diuretic hormone receptor 1 (Dh44R1) in pC1 neurons almost completely abolished cAMP elevation induced by sine song. Thus, I propose that Dh44 signaling in pC1 plays a role in the acoustic priming effect of female reproductive behavior in the fruit fly
