Simple paper-based air sampling and DNA extraction kit for molecular diagnosis of airborne pathogen

- In order to defense bioterrorism or prevent pandemic diseases, the requirement for detection and monitoring of airborne pathogens has gradually increased. Despite that, complex instruments and expensive costs for transport are still needed. Here, we developed the simple paper kit for airborne pathogen monitoring. Air collection and PCR analysis of airborne pathogens were simply conducted with the operation of air pumps in manufactured biosafety cabinet systems. The bacteria collected in the paper kit were washed, then the DNA of the bacterial cells was extracted with a micropipette to quantify the airborne bacteria. Components of the paper kit and condition of pump operation were optimized for airborne bacteria in the biosafety cabinet. Using the paper kit, airborne bacteria of approximately 3000 CFU / m3 were collected and within 30 minutes from the concentration of nebulizer solution of 2.4 x 105 CFU / mL. This paper kit and technique could be a useful alternative for purpose in simple, rapid, and cost-effective airborne bacteria monitoring system. We expect this study to contribute to the detection of airborne pathogens.
