A fluorogenic peroxidase substrate based on hydrazone and its applications in detection of glucose and cysteine

Abstract
Peroxidase is increasing in interest because it has a wide potential analytical application area. However, current peroxidase substrates for measuring peroxidase activity has limitation in cost, stability and overoxidation. In this work, we developed a fluorogenic peroxidase substrate based on hydrazone moiety. This fluorogenic substrate oxidized upon addition of H2O2 under horseradish peroxidase at acidic conditions. Based on these results, this substrate was applied to detection of glucose with enzyme cascade reaction of horseradish peroxidase and glucose oxidase. For the further application, it was applied to measurement of peroxidase-like activity of gold nanoparticles and detection of cysteine with gold nanoparticles.
