- Development of a microfluidic based antibody microarray chip for the detection of multiple biomarkers
- Abstract
- The microfluidic-based antibody microarray has advantages such as low volume requirement, multiplexed detection. This microarray can be applied to various research as much as cancer research. In this study, the antibody microarray chip is developed to detect cardiovascular disease (CVD) related multi-biomarker. To fabricated microarray chip, antibodies are immobilized on a slide glass by surface modification with a chemical. All samples are injected into the microfluidic channel and incubated. The targets are detected by fluorescence intensity through antibody-antigen binding and fluorescence-labeled streptavidin-biotin binding. Fluorescence intensity increase as the target concentration increase. Finally, the developed microarray was evaluated by simultaneously detecting MCP-1, IL-6, and TNF-α related to CVD. As a result, a graph showed that as the target concentration increased, the fluorescence intensity increased. The logistic model was used to analyze the fluorescence intensity of MCP-1, IL-6, and TNF-α. It showed R2 are 0.88, 0.98, and 0.76, respectively.
