- Abstract
- Sepsis is caused by infection with bacteria and a severe inflammatory reaction appears throughout the body. If this sepsis is not treated early, the mortality rate reaches 40%, so prompt diagnosis and treatment are important. However, blood consists of more than 5 billion blood cells per ml, so it is very difficult to separate extremely low concentrations of bacteria (1-100 CFU/ml). Therefore, in this study, we report a hemocyte lysis and bacterial separation chip based on joule heating. Bacteria, unlike blood cells, have capsules made up of polysaccharides, making them resistant to heat. Therefore, when the blood is heated, the blood cells are lysed and the remaining bacteria can be separated. In order to confirm the performance of chip, blood diluted 20 times with PBS (Gibco 10x, pH7.4) was used. (Escherichia coli, 50~100CFU/ml) Also, The Inlet flowrate was 700ul/h, DC power 12V was applied. As a result of the experiment, the blood cell lysis efficiency was 99.9% and the bacteria separation efficiency was 68.8%(N=3). These results show that it is possible to isolate low concentrations of bacteria in the blood containing many blood cells. The separated bacteria can then be rapidly identified by antimicrobial-susceptibility technology, which is expected to enable rapid diagnosis and treatment of sepsis.
