Identification of immune-regulating gut microbiota in mice skin graft model
- Abstract
- The composition of microbiota has important correlation with various skin diseases. Not only skin microbiota manipulates it, but also astonishingly a lot of skin disorders are influenced by alteration of gut microbiota through gut-skin axis. The organization of gut microbiota is a crucial factor of our immune systems in body. But, a little number of studies have researched the possibility of gut microbiota to improve the status of affected area of skin transplantation. The purpose of the current study is to research whether probiotics which can regulate skin immune reaction exist and treatment of these probiotics can enhance the progression of skin transplantation. Human keratinocyte cells was cultured with lipopolysaccharide (LPS) for inflammatory stimulation and supernatant of culture media of Lactobacillus (L) rhamnosus, L. salivarius is added to human keratinocyte cells. By using WST-8 assay, L. rhamnosus, and L. salivarius significantly increased the recovery rate of cell viability in cells damaged by LPS. Also, L. rhamnosus has suppressed the mRNA expression of immune-activating genes : IL-6, TNF-α whereas L. rhamnosus has increased the mRNA expression of immune-suppressing genes : IL-10, TGF-β. Mouse group of graft model that had oral administration of L. rhamnosus showed elevated graft survival rate. And, the benefit of immunosuppressive drugs has synergic effect with L. rhamnosus administration in mouse graft model. Therefore, these data suggest that L. rhamnosus has potential to excelerate recovery of skin transplantation in terms of immune regulation.
