Deep proteome analysis of two edible insect species, Gryllus bimaculatus and Protaetia brevitarsis using high resolution LC-MS/MS
- Abstract
- Edible insects have been proposed as a new substitute for food shortages due to a growing population. The inclusion of insect in food should be strictly regulated as it can cause food allergies and anaphylaxis. Therefore, analytical methods capable of distinguishing and discriminating each insect species in food are essential. In this study, protein extraction efficiency was compared for each type of extraction buffer prior to the analysis of edible insect proteins. Then, species-specific marker peptides were identified for two species of insects, two-spotted cricket (Gryllus bimaculatus) and white-spotted flower chafer larvae (Protaetia brevitarsis), with high protein and unsaturated fatty acid content. Finally, it was investigated whether they were detected without being denatured in processed food. As a result, urea and hexane showed the best extraction efficiency, and these were used for LC-HRMS analysis. Also, it was identified that 14 species-specific marker peptides of the two-spotted cricket and of the white-spotted flower chafer larvae through BLAST. These marker peptides were also detected in processed foods, confirming that they were not affected by the processing method.
